As much as I hate to say it, I think Hercule (Mr. Satan to use his name from the original Japanese version) would have the advantage since he’s the only one of the two with any actual training and experience in combat, such as it is. Not only that, but Mr. Satan usually has a weapon hidden somewhere on his person whereas Iron Will is unarmed unless you count his horns.

Keep in mind Hercule (Satan, whatever) was a joke only compared to the planet-crushing main characters and villains, in any other series he would have been an unstoppable beast. Even in the world of FiM his sheer strength is insane.
