4th cousins means they have the same great-great-great-grandparents, 5th means one more then that, “removed” means not of the same generation with “twice” meaning it’s two generations.  
So my aunt’s daughter is my first cousins and, her son is my first cousins once removed.

Thanks for the reply. I still have 2 questions though:
 
Q1:  
What did she mean by “have a 4th cousin twice removed ‘by’ a 5th cousin”?  
Can I replace that “by” with “or”?
 
Q2:  
Applesauce is a 4th cousin twice removed of the Pie family.  
But to whom from the Pie family?  
I can’t help but wonder how Pinkie concluded that she is AJ’s 4th cousin twice removed with that given statement.
 
 
Either I’m derping, or Pinkie’s just being Pinkie Pie. :S

Fourth cousin twice removed would result from one of two instances.  
  •  
    Pinkie being the 3rd great granddaughter and Applejack being the 5th great granddaughter of their shared ancestor  
    -or-  
    Pinkie being the 5th great granddaughter and Applejack being the 3rd great granddaughter.
