I have to say the best overall casting choice would be Twilight Sparkle.  
Pinkie would have been great for Star first season but not as great as Star develops over time.  
Starlight would be a poor man’s Twilight in of casting.  
Anypony else isn’t really in the running unless you aged up Flurry Heart and took advantage of that blank slate.

Some sessions I feel so alone I could cry, but I don't. I never do. Because what would be the point. Not a single person in the entire ladder would care. Take it to your grave.

I’m now imagining Spike talking to her in a deep voice. Yes, Spike is Marco in this scenario.  
Rarity would be Jackie, but that’s not as relevant.  
Smoulder would be Janna and that’s virtually irrelevant.  
Luna as Eclipsa is totally irrelevant but amusing to me.
